[ Anotasi Abstrak ]
Deskripsi Lain
Gili Labak Island is an area that has a good coral reef ecosystem, including the biota that live in it. Sponge is one group of biota that has the potential to produce secondary metabolites and has bioactive properties. Sponges can accumulate bacteria and fungi in the body. According to several studies showing that bioactive compounds can be found in Demospongia class sponges. New antibiotic compounds are very useful for fighting pathogenic bacteria that have been resistant to several antibiotics. The purpose of this study was to determine the potential of spongy symbion fungi that were able to fight pathogenic bacteria Staphylociccus aureus and Escherichia coli. Sponge samples were taken from the waters of Gili Labak island, Sumenep, East Java. There were 2 samples of sponge that were successfully isolated. Symbion fungal isolation uses PDA media. The Symbion fungal purification uses the scratch method. The purification results obtained 5 fungal isolates from 2 sponge samples. The preliminary test was carried out by the diffusion method so that the one who succeeded was the symbion fungus isolate coded GL-S2-02. Next is the fungal culture process. After that the extraction process is carried out. Then the mushroom extract test was carried out by disc diffusion method. The test results showed that the Demospongia class sponge symbion fungal extract (Aaptos aaptos) with a concentration of 250 μg/disk was able to inhibit the growth of pathogenic bacteria with an average value of 3.5 mm E. coli and 3.2 mm of S.aureus. The results of the molecular identification of fungi showed that the type of active fungal isolate was Penicillium citrinium. The conclusion of the study is that fungal isolates from sponge Aaptos aaptos have the potential to produce bioactive compounds to fight bacteria that have been resistant to several antibiotics.
