[ Anotasi Abstrak ]

Deskripsi Lain

Mycoviruses are viruses that infect fungi and have the potential to influence the biological characteristics of their hosts, including virulence. One type of fungus that can serve a host for mycoviruses is Fusarium sp. This study aims to determine the ability of four specific primer pairs—FgV1, FcPV1, FcMV1, and FcMOV1—to detect mycovirus targets in Fusarium sp. isolates from various sources. The study was conducted a descriptive-exploratory study using a molecular approach at the Center for Development of Advanced Sciences and Technology (CDAST), University of Jember, from October 2025 to August 2026. The study utilized 11 Fusarium sp. isolates sourced from various crop commodities and locations. The research stages included total RNA isolation, enzymatic treatment using S1 nuclease for dsRNA enrichment, complementary DNA (cDNA) synthesis, amplification using the RT-PCR method with four specific primer pairs tested separately, and analysis of the amplification products via 2% agarose gel electrophoresis. The isolation results showed that total RNA concentrations ranged from 119.76 to 1,265.00 ng/µL, with A260/A280 ratios of 1.997–2.212 and A260/A230 ratios of 0.884–2.440. The electrophoresis results showed that no DNA bands at the expected target sizes were detected in any of the 11 isolates for all four primer pairs used. Thus, the mycovirus targets examined were not detected under the RT-PCR conditions applied. These results cannot be used to conclude that all isolates are free of mycoviruses because detection was limited to the four targets tested. Further research is needed using positive controls, evaluation of RT-PCR conditions, and additional primer pairs to expand the scope of mycovirus detection in Fusarium sp.
